POS0363 IDENTIFICATION OF MOLECULAR PHENOTYPES AND IMMUNE CELL INFILTRATION IN PSORIATIC ARTHRITIS PATIENTS’ SKIN TISSUES BY INTEGRATED BIOINFORMATICS ANALYSIS

Background: Psoriatic arthritis (PsA) is an inflammatory musculoskeletal disease associated with cutaneous psoriasis 1 . Heterogeneity of clinical manifestation often makes differential diagnosis difficult 2 Thus, the underlying molecular pathogenesis PsA need to be further studied diagnose early and ensure optimal management key comorbidities. Objectives: This research was conducted identify phenotypes immune infiltration in skin tissues psoriatic patients according bioinformatics analysis. Methods: The mRNA expression profiles GSE13355 (116 samples), GSE14905 (56 samples) GSE30999 (162 were obtained from publicly GEO databases. Non-negative matrix factorization (NMF), functional enrichment cibersort algorithm applied illustrate conditions patients’ for classification after screening differentially expressed genes (DEGs) between lesion biopsy non-lesion biopsy. Results: Two subsets (Sub1 Sub2) identified validated by NMF typing 612 detected DEGs (Figure 1a). A total 54 signature (18 Sub1 36 1b). GO KEGG analysis showed mainly involved proliferation differentiation cells, whereas Sub2 related humoral response mediated antimicrobial peptide 1c.1d). Further, cell results revealed had higher levels resting NK cells ( P <0.001), macrophages M1( mast <0.001) regulatory T but lower concentrations activated CD4 + memory <0.05), dendritric cells( eosinophils <0.05) neutrophil 1e).
